U.S. Route 16 (US 16) is an east-west US Highway in central and northern Wyoming in American Truck Simulator. The route is separated into two sections: The first section runs concurrent with US 14 and US 20 from US 89, US 191, and US 287 in Yellowstone National Park to WYO 789 in Greybull, WY. The second section runs concurrent with US 14 from WYO 59 in Gillette to I-90, where the three routes travel east to Moorcroft, WY before separating. In real life, US 16 runs 540 miles (870 km) between Yellowstone National Park and Rapid City, South Dakota.
Course[]
US 16 enters Greybull inaccessibly from the north with US 14, US 20, and WYO 789. US 16 then continues south toward Thermopolis in concurrence with US 20 and WYO 789 for a few miles before the road turns into just US 20 and WYO 789.
US 16 reappears in Gillette, running east in concurrence with US 14 from a Steeler depot where it travels through downtown Gillette to I-90 at exit 128. US 14 and US 16 then join I-90 and travel east to Moorcroft, where they split from I-90 at exit 154. US 14 and US 16 then travel south briefly before US 16 splits off and travels east, inaccessibly toward Upton and Newcastle.

- In the initial version of the Wyoming map expansion DLC, the section of US 16 concurrent with US 14 from WYO 59 in Gillette to Moorcroft was added.
- In update 1.45, the section of concurrent with US 14 and US 20 from US 89, US 191, and US 287 in Yellowstone National Park to WYO 789 in Greybull was added, along with Cody.
